Yes, you can. It's not hard. Basically all you do is setup FBO's and make sure the freight will fit legally and safely. It varries on the operation though. Sometimes pilots will have you file for them and other times pilots will do it. Part 135 Followers DO NOT have operational control unless the D.O. gives permission to you. The other thing is it's whatever the pilot says goes...unless the D.O. interveens.

As for the endorcement for ADX, it's exactly what Twin Wasp said. If you know what you're doing, it's easy and quick. Just know duty times and study up a bit on the 727-200.
