Keep trying. In my own experience it doesn't seem like Skywest looks down upon CFIs like other regionals do. My new hire class had 12 CFIs all with no turbine time and all 12 of us passed everything first try. A lot of the captains I fly with say they were CFIs before being hired by Skywest.
They care a lot more about who you are and how you conduct yourself in the interview and how well you prepare for the interview.
I'll second that, I was a CFI hired strait into the jet last march. From my experience the biggest factors are getting the interview and knocking it out of the park. As far as I know only 3 out of the 15 of my interview group got hired (one being a rehire) and almost everyone except me had 121 experience.
