It's not based upon a duty day but a duty period; a period can cover multiple days. A CDO/Stand-Up is one duty period as is a single day of flying. The 3:45 is a min guarantee per duty period/day, above that you are guaranteed at least half your duty period and above 12 hours you get 6 (12/2=6) hours plus 1:1 for every minute scheduled over 12 hours. This is all based upon a "scheduled period/day" and doesn't change if your day changes, that's why it's a guarantee; the only time this is not true is when you block more.
Example:
Scheduled to Fly:
13 hour duty period/day with 5 hours of block is credited at 7 hours of pay. (6+1=7)
Actual:
14:30 hour duty period/day with 8:30 hours of actual block pays 9:30. (8:30+1=9:30)
