my advice would be to get to all the mins for an atp as quickly as you can and by the time you do the regionals will be hiring anyone they can. If you can get the required night and XC time instructing as a regular cfi that’s great, if you can’t get your cfii. I figure it’ll take you about 2 years to get to 1500 and your multi time depending on how much you instruct. I hated instructing so I went the 135 route and I think it made me a lot better pilot. Once you get to the regionals it doesn’t matter how you got there so long as you pass check-rides. So far as major airlines wanting people who flew 135 vs instructed I have no idea, but at a regional all qualified applicants will soon be welcomed.
