Default PT job to pilot job?

Assuming you meet the hiring requirements, would getting a part-time job at major airline increase your chances of getting an interview there as a pilot?

In a very general sense, yes, but not always. If you want to increase your odds, try to get a job in flight ops, or where you will have access to a Chief Pilot.

Most airlines provide their own employees an inside track, but being part-time may not be enough.

If you do get the PT job, prepare to absolutely bust your tail to make the right impression, never say anything negative about the company, always show up early and stay late, and make it clear what your goal is.
