Ops Specs DO supercede regs (like allowing take offs at below standard minimums.) But since most 135 (or 121) Ops Specs are pretty much boilerplate, I'll bet you would have a hard time finding one that supercedes rest requirements. Because it would have had to be specifically signed off by an FAA representative. Wouldn't happen.
