For example, I have worked for Delta & Comair in the past in a non-flying capacity. If I were hired by Chautauqua or Skywest, or one of the other "Delta Connection" carriers, can I request to fly the "Delta Connection" line/routes?

SkyWest - Eventually you could...but only by bidding into SLC or ATL for the Delta flying. This doesn't mean that you won't flow-through DEN, ORD, or Cali, on the United-side once in awhile...
In this industry your priorities are probably better off focusing on something other than the livery and color-scheme on the plane you're flying LoL - you're going wherever the Airline feels it's best for
