No, you only get the 5:15 if the day of vaccine + 48 hour waiting period is all on days off. If they have to drop any trip or reserve days over that 3 day period you get the drops paid but no extra 5:15.

For your scheduling, it's 3 calendar days off. One for the shot and 2 for the 48 hour down period.

You could possibly work it out to only using 2 off days if you could get the shot either at your base or at home immediately following the last leg of a trip.
