Back in the day SkyWest used to do a EAS route of Visalia to Fresno in a Brasilia, which are 32 nm apart. The goal was to try to break 6 minutes of flight time.

Also used to do Ontario to LAX. A little longer (41nm) but I remember the clearance when departing westbound was "Fly runway heading and intercept the LAX 24R localizer."

Looks like we have a winner for routes currently flown. Cape's shortest part 121 route is GUM-ROP at 49NM.

DEN-COS is pretty short at 63 nm. That's very quick, especially departing/landing south.

We used to fly MIA-FLL (23 miles) in the 727. It followed a late-night ATL-MIA leg and usually had only a few pax.

13 minutes for the reverse route in a 757.

I've done OAK-SFO at FedEx. It used to be done often to reposition the plane.
Took off on 11 and landed on 28R, 6 minutes airborne in an MD-11.

No idea, this company seems to make money even doing odd things sometimes. I flew a empty B777 form MEM to ICN once.
