I'm in the process of amending our ops specs and while I'm almost certain we don't do supplemental operations, curiosity is begging the question, What the heck are supplemental operations? Any knoweldgeable explanation would be appreciated.

A lot of the 121 cargo operators fly on supplemental certificates. The main differences are no true dispatchers and some duty time regs may be different. The pic pretty much bears all the responsibility for the flight as opposed to flag operations in which it is a shared responsibility with the dispatcher and pic.

The big differences were you always had a filed alternate which meant you always carried the extra fuel. No 7 day flight time limits so you could fly 85 hours in two weeks and go home. Rolling 30 day and 12 month flight time limits v. monthly and yearly. As Rama said, Flight followers v. Dispatchers (even though our POI required them to have Dispatcher certificates. It's the DO and PIC who agree the flight can proceed and the Flight Followers are the DO'd agents.
