Leadership boxes can be checked regardless of platform. Your best bet is to plan on a purely civilian timeline, but if I had to guess it could be quicker depending on the company.

Looks like JetBlue may be a good stepping stone on the way as they do take full rotor time as well

Certainly this has been true for a long time. But is that changing?
There are a lot of newly hired mainline FOs who don't have any TPIC. I know a few myself.
With the looming retirement wave, it increasingly appears as though 121 time...and total time...are the big items. Certainly TPIC doesn't hurt. But in the future I don't think the majors are going to pass on a guy who has 4-5K hrs with 2-3K 121 time, has no busts (legal or training-wise), a college degree, speaks and presents him/herself well, has internal recs, is in their 30s or early 40s (read: stable, married, kids, no divorces), and as good in an interview as he/she is on paper.
They have 20,000K more jobs to fill in the next 8-10 years.

1 - The right kind of resume fluffing.
2 - Internal help.
2a - sometimes a pinch of demographics to mix things up.
Same as every other industry.
