It depends, without looking at all their websites, I'm sure they would accept it for total time. Landing uses all the same skills as a landplane, plus some additional ones.
They would probably accept it for PIC/TPIC also, but I think if your TPIC was all seaplane ops, you would be at a competetive disadvantage...
I can't think of too many really large seaplanes in service, some major airlines like experience in RJ-size or better.
The nature of the operations would play a role, if it was a 121/scheduled 135 that might be OK, but if it was a whole lot of single-pilot VFR they would prefer someone with experience in the IFR system and a crew cockpit.
For regionals it should be fine, but read their web pages for detailed hiring requirements.

That's stick-and-rudder intensive flying, but will not give you the crew/IFR experience. If you do fly IFR, it will be different from 121 IFR.
I wouldn't bet on making the jump from twotters to a major, maybe not impossible but probably not likely. I think Alaska might hire some bush pilots, but not sure.

I wasn't planning to build all my "pre-major" time there- more of a time-building deal.
FDX and UPS want time in planes weighing 300,000lb?! That's prpbably around the weight of an A310. Are you saying you'd have to leave a major to get into those 2 companies?

They have plenty of ex-major pilots...they pay twice as much if you are low-seniority. I heard DAL was only hiring guys with slightly less than 1000 TPIC for a while because too many of their new hires were bailing for UPS.
